Castlemaine is an important stop on the line to Bendigo. It is also the junction of the cross country line to Maryborough. The line arrived in Castlemaine by 1862, and a contract was let for the construction of the first signal box in 1888. By 1915 it was replaced by the current structure on the island platform. The box (and most of the semaphore signals) were abolished in 1989, replaced by a panel in the station building and coloured light three position signalling. However the box has been given a new lease of life since 2004, controlling the signals and sidings for the Victorian Goldfields Railway, who use No 3 platform at the station and some of the old yards to the North of the Station.

The cross country line was closed between Castlemaine and Mooloort in 2004 to allow the VGR to operate the line between Castlemaine and Maldon Junction.
